如何在Geneious Prime中添加引物

引物是Geneious Prime中的一种特殊文件类型。引物文件将带有绿色箭头图标在文档表中。  A primer normally comprises a binding region, which is denoted by the “Binding region” annotation, and optional 5’ extension which may include adaptor sequence, restriction sites or a combination of elements.  

当将引物添加到Geneious中时，重要的是要确保将它们格式化为引物文件，否则它们将无法用于与引物相关的操作（例如针对新序列进行测试）。

本文介绍了如何通过手动输入或从外部来源导入将现有引物导入Geneious Prime的方法。 

1手动输入各个引物序列

If you want to enter a primer sequence, for example a published primer sequence, go to File → New → Sequence.这将打开“新序列”窗口，只需输入引物序列并将“类型”设置为“引物”即可。如果引物具有延伸，请选择与结合区域相对应的区域，然后单击“结合区域：”按钮以设置结合区域起点。

单击确定，将创建新的底漆文件。

或者，您可以在创建引物之后添加扩展名。为此，请按上述步骤创建一个引物，但不包括扩展名。Once the primer is created, select the new primer and go Primers → Add 5′ Extension.这将允许您将带注释的扩展元素（例如限制位点或网关位点）添加到引物中。

2从电子表格或带分隔符的文本文件中导入多个入门。

您可以使用“ 文件”菜单下的“ 导入”选项，或者从文件中复制和粘贴，轻松地从电子表格或带分隔符的文本文档中导入引物序列。At a minimum the tabular data should have two columns, the first column for the primer name, the second for the primer sequence (5′-3′).

To import from the File menu… If you have a collection of primers in an Excel spreadsheet then you can save the file in either tab-delimited (.tsv) or comma-delimited (.csv) format.Then either import via the menu File → From File…, or simply drag the delimited file into the Geneious Prime window.Geneious将识别您正在导入表格数据，并要求您确认导入格式。确认格式后，将打开“导入序列”窗口。  

要通过复制和粘贴导入...如果您在Excel电子表格或带分隔符的文本文件中有引物，则可以简单地从电子表格中复制（CTRL-C或Command-C）列数据，转到Geneious Prime and Paste（CTRL-v或Command-v）。Geneious  will detect that you are importing tabular delimited data and ask you to confirm the import format.确认格式后，将打开“导入序列”窗口。

The Import Sequences window, shown in the screenshot below, displays a preview of how the sequences will be imported into Geneious.确保将类型设置为Primer，并配置字段，以便将Name，Sequence，Description和Primer Extension（如果需要）设置为定界文件中正确的列（剪贴板内容框显示文件中包含的列）。  请注意，如果您有引物扩展名，则必须在文本文件中绑定区域的单独列中列出引物扩展名，以便可以正确识别它们。然后单击“确定”完成导入。

3将引物添加到序列中。

当您要测试已发布的引物序列（当前不在Geneious数据库中）是否与数据库中的特定序列匹配时，应使用 “ 引物” 菜单 下的此操作 。 只需输入或粘贴序列，配置所需的任何不匹配余量，然后单击“确定”。

如果找到匹配项，则引物将在您的序列上标注，然后您可以根据需要将其提取到数据库中。  

注意，该操作目前仅适用于不带引物的引物。
